Principle | Proteose peptone and yeast extract supply nitrogenous compounds and other nutrients. Sodium thioglycollate ensures anaerobic conditions. Disodium phosphate acts as a buffer. Raffinose is specifically fermented by C. perfringens to produce acid within 72 hours. Acid production is tested by adding bromothymol blue, turning the medium yellow. |

Directions | Add 34 g to 1000 ml distilled water and mix well. Heat if necessary to dissolve the medium completely. Autoclave at 15 pounds pressure (121°C) for 15 minutes. Dispense into sterile tubes. Check one or both tubes to measure pH. |
