Summary | The enzyme reversibly acts on vicinal hydroxyhalides (vicinal halohydrins) to form epoxides and open the epoxides, which can be used for the synthesis of various chiral pharmaceuticals and fine chemicals. |
Description | Halohydrin Dehalogenase is produced by submerged fermentation followed by separation, ultrafiltration and formulation. The enzyme reversibly acts on vicinal hydroxyhalides (vicinal halohydrins) to form epoxides and open the epoxides. |
Mechanism | Halohydrin Dehalogenase catalyzes the chemoenzymatic removal of halogens from vicinal halohydrins, generating chiral epoxides through an intramolecular nucleophilic substitution mechanism. It can catalyze the ring-opening reaction of epoxides in the presence of nucleophilic reagents such as CN-, N3-, SCN-, and OCN-, producing chiral β-substituted alcohols, and forming new C-N, C-C, and C-O bonds. Chiral epoxides and β-substituted alcohols are key chiral building blocks for the synthesis of various chiral pharmaceuticals and fine chemicals, with broad application prospects. |
Advantages | Removal of halogens from vicinal halohydrins; catalyze the ring-opening reaction of epoxides in the presence of nucleophilic reagents; the synthesis of various chiral pharmaceuticals and fine chemicals |
