Overview

Summary The enzyme reversibly acts on vicinal hydroxyhalides (vicinal halohydrins) to form epoxides and open the epoxides, which can be used for the synthesis of various chiral pharmaceuticals and fine chemicals.
Description Halohydrin Dehalogenase is produced by submerged fermentation followed by separation, ultrafiltration and formulation. The enzyme reversibly acts on vicinal hydroxyhalides (vicinal halohydrins) to form epoxides and open the epoxides.
Mechanism Halohydrin Dehalogenase catalyzes the chemoenzymatic removal of halogens from vicinal halohydrins, generating chiral epoxides through an intramolecular nucleophilic substitution mechanism. It can catalyze the ring-opening reaction of epoxides in the presence of nucleophilic reagents such as CN-, N3-, SCN-, and OCN-, producing chiral β-substituted alcohols, and forming new C-N, C-C, and C-O bonds. Chiral epoxides and β-substituted alcohols are key chiral building blocks for the synthesis of various chiral pharmaceuticals and fine chemicals, with broad application prospects.
