Description Glucosamine-2-epimerase is produced by submerged fermentation followed by separation, ultrafiltration and formulation. The enzyme catalyzes the interconversion between N-acyl-D-glucosamine and N-acyl-D-mannosamine and is a key enzyme in the synthesis of N-acetyl-D-neuraminic acid, which is a precursor for the synthesis of other sialic acid substances.
Mechanism In the enzymatic synthesis of N-acetylneuraminic acid, the first step is the isomerization of N-acetyl-D-glucosamine to N-acetyl-D-mannosamine by N-acetylglucosamine-2-epimerase. Subsequently, N-acetyl-D-mannosamine condenses with pyruvic acid under the action of N-acetylneuraminate lyase to form N-acetylneuraminic acid.
