Principle | This test is similar to oxidation-fermentation tests for non-fermentative organisms. Tryptone and yeast extract provide essential nitrogenous nutrients, while glucose serves as the fermentable carbohydrate. Bromo cresol purple is the pH indicator. A small amount of agar creates anaerobic conditions in the tube depths. |

Directions | Suspend 23.24 g in 1000 ml of purified/distilled water. Heat to boiling to dissolve the medium completely. Dispense in tubes and sterilize by autoclaving at 15 pounds pressure (121°C) for 15 minutes. Allow the tubes of medium to cool in the upright position. |
