Product Name Fermentation Medium for Neisseriae
Aspect Light yellow to light pink powder
Application | The product is used to study the fermentation reactions of Neisseria. |
Principle | Tryptone provides necessary nitrogenous nutrients. L-Cystine acts as both an amino acid source and a reducing agent, removing molecular oxygen to prevent peroxide accumulation. Agar reduces convection currents, maintaining anaerobic conditions. Sodium chloride preserves osmotic balance. Phenol red, turning yellow at acidic pH, indicates carbohydrate oxidation. Pink color indicates peptone degradation. Neisseria species produce acids near the inoculated area, causing potential medium-wide yellowing if contaminants are present. |
pH | 7.5±0.1 |
Colour and Clarity of Prepared Medium | Straw, clear to slightly milky gel |
Formulation (/L) |
Tryptone 20.0 g L-Cystine 0.50 g Sodium chloride 5.0 g Sodium sulphite 0.50 g Phenol red 0.0170 g Agar 3.50 g |
Directions | Suspend 29.52 g in 1000 ml of purified/distilled water. Heat to boiling to completely dissolve the medium. Autoclave at 118°C for 15 minutes. Pressure should not exceed 12 lbs. Cool to about 40-45°C and add membrane filter sterilized sugar solution to a final concentration of 1%. |
