| Directions | Suspend 22.25 g in 1000 ml of purified/distilled water. Heat to boiling to completely dissolve the medium. Dispense 9 ml into 16 X 125 mm test tubes containing inverted Durham tubes. Autoclave at 15 pounds pressure (121°C) for 15 minutes. Prior to use, the tubes were heated in boiling water or free-flowing steam for 10 minutes to remove dissolved oxygen and 1 ml of 1% sterile carbohydrates-salicylic acid and cotton-cellulose-are added to separate tubes. |
