Description Ethidium homodimer-I (EthD-I) is a high affinity fluorescent nucleic acid dye that enhances fluorescence more than 30-fold when combined with DNA or RNA. It is used for staining mammals, bacteria, yeast and fungi. EthD-I has a strong positive charge, so the dye does not penetrate cell membranes for live cell staining, but it does penetrate the disordered regions of dead cell membranes to reach the nucleus and embed itself in the DNA double strand to produce a red fluorescence. Therefore, EthD-I is a sensitive nucleic acid stain that can accurately detect nucleic acids in solution or in disintegrating cells.
